RPMI 1640 is a medium that was developed as a modification of McCoy’s 5A medium and is based on RPMI 1630. The RPMI 1640 uses a bicarbonate buffering system and it differs from other mammalian cell culture media as it has a typical pH 8 formulation. RPMI 1640 with or without supplemented serums or defined proteins, is the most widely applied culture medium for short- and long-term cultivation in many cell types, especially human T/B-lymphocytes, bone marrow cells and hybridoma cells.
